Therefore, we have been devel- oping another molecular technique (LAMP) that could be used by growers at their desk or will help reduce the cost of a service. Over two years, the results were very similar to the qPCR standard, and the management decision resulted in similar dis- ease levels in the vineyard. However, there was ambiguity in visual assessment of the positive and negative results that resulted in false reading. The results were also only qualitative and not suitable to help growers use the detection data to adjust spray intervals after initiation of a fungicide program. In order to use LAMP to quantify spore concentration and improve its accuracy, we partnered with Diagenetix Inc. to develop a quantitative LAMP assay using their hand-held SMART-Dart platform and Assimilating Probe technology. In 2012-13, we have shown that the Assimilating Probe technology is extremely specific and sensitive for detecting and quantifying E. necator. In 2014, the SMART-Dart will be tested by growers. Currently, we can say a sample contains zero, less than 10, or accurately estimate spore concentration from 10 to 10,000 spores. The newest iteration of the SMART-Dart device will be commer- cially available in 2014.
